Regulation of ferritin synthesis in rat liver.

چکیده
Ferritin synthesis in rat liver is greatly increased by prior treatment of intact animals with iron salts. Liver slices from treated rats incorporate increased amounts of Wleucine into ferritin in vitro, with net synthesis of the protein. Slices of spleen, testis, intestine, and kidney also show increased incorporation following iron. The response of the liver to iron has been studied to determine whether increased transcription of genes and the formation of messenger RNA are involved, or whether control is at a later stage in the biosynthetic pathway. Three lines of evidence support the existence of transcriptional control. (a) Actinomycin D inhibits the response to iron. (b) Iron treatment stimulates incorporation of 14C-orotic acid into a rapidly labeled fraction of liver nuclear RNA. (c) Direct addition of iron to liver slices in vitro has failed to stimulate ferritin synthesis, although jgFe penetrates liver cells.

Biosynthesis of Ferritin by Rat Liver Slices.
Iron greatly accelerates the biosynthesis of the protein moiety of the iron storage protein ferritin (1, 2). This phenomenon is therefore of general interest as an example of the adaptive control of specific protein synthesis. As a possible tool for studying the process in vitro, rat liver slices were tested for their ability to incorporate labeled amino acids into ferritin. In this report we d...
متن کاملTranslation of ferritin light and heavy subunit mRNAs is regulated by intracellular chelatable iron levels in rat hepatoma cells.
Acute administration of iron to rats has been previously shown to induce liver ferritin synthesis by increasing the translation of inactive cytoplasmic ferritin mRNAs for both heavy (H) and light (L) subunits by mobilizing them onto polyribosomes. In this report rat hepatoma cells in culture are used to explore the relationship of this response to intracellular iron levels. After adding iron as...
متن کاملIdentification and characterization of a receptor for tissue ferritin on activated rat lipocytes.
Hepatic iron overload causes lipocyte activation with resultant fibrogenesis. This study examines whether rat lipocytes express ferritin receptors, which could be involved in paracellular iron movement and in cellular regulation. Lipocytes from normal rat liver were cultured on plastic and incubated with 125I-labeled rat liver ferritin (RLF) +/- a 100-fold excess of either unlabeled RLF or huma...
متن کاملBoth subunits of rat liver ferritin are regulated at a translational level by iron induction.
A few hours after administering iron to rats, liver ferritin synthesis increases several fold. However, Northern blot analysis with cDNA probes for ferritin light (L) and heavy (H) subunit mRNAs failed to show an increase in total population of either messenger. Cytoplasmic distribution of ferritin messages was therefore investigated in control and iron administered rats killed at 3.5 hours. Th...
